Tau-targeting therapy in Alzheimer’s disease: critical advances and future opportunities
نویسندگان
چکیده
Alzheimer’s disease (AD) is a progressive neurodegenerative disorder characterized by two pathological hallmark lesions: extracellular plaques composed of ?-amyloid (A?) peptide and intracellular neurofibrillary tangles made up highly phosphorylated tau protein. Over the past decades, most disease-modifying therapies against AD have been developed mainly on basis amyloid cascade hypothesis with focus A?. However, these agents yielded only limited benefits progression, which prompts us to revitalize long-neglected hypothesis. Tau protein microtubule-associated protein, can stabilize microtubules, regulate microtubule assembly, affect morphology growth neuronal axons. Much more importantly, degree pathology closely related cognitive decline in patients than that A? pathology. Therefore, tau-targeting therapy seems be promising approach combat AD. This review describes research progress AD, an emphasis immunotherapy. The current challenges future perspectives this field are also discussed.

Barriers and recent advances in non-viral vectors targeting the lungs for cystic fibrosis gene therapy
Cystic fibrosis (CF) is an autosomal recessive disorder caused by mutations in CFTR genes that affect chloride ion channel. The CF is a good nominee for gene therapy as the asymptomatic carriers are phenotypically normal, and the desired cells are accessible for vector delivery. Gene therapy shows promising effects involving the correction of gene or replacement of the mutant gene with the func...
متن کاملResearch opportunities and advances in lung disease.
Lung diseases cause morbidity, mortality, and economic burden. Susceptibility to pathogenesis of most lung diseases result from a complex interaction of a relevant challenge from the environment, genetic background, and the nature of the host responses. Cutting-edge research in lung disease now centers on understanding the lung as a genetically determined biological organ that functions to medi...
متن کاملAdvances and Future Challenges in Adenoviral Vector Pharmacology and Targeting
Adenovirus is a robust vector for therapeutic applications, but its use is limited by our understanding of its complex in vivo pharmacology. In this review we describe the necessity of identifying its natural, widespread, and multifaceted interactions with the host since this information will be crucial for efficiently redirecting virus into target cells. In the rational design of vectors, the ...
متن کاملImage-based musculoskeletal modeling: applications, advances, and future opportunities.
Computer models of the musculoskeletal system are broadly used to study the mechanisms of musculoskeletal disorders and to simulate surgical treatments. Musculoskeletal models have historically been created based on data derived in anatomical and biomechanical studies of cadaveric specimens. MRI offers an abundance of novel methods for acquisition of data from living subjects and is revolutioni...
متن کاملRecent Advances in Hemophilia Gene Therapy
Heterogeneous loss of function mutations at F8 and F9 genes causes X-linked recessive bleeding disorders, hemophilia A (HA) and hemophilia B (HB), respectively. HA is clinically indistinguishable from HB and accounts for more than 80% of hemophilia cases; the former affects 1/5000 and the latter 1/25000 male births worldwide. In Iran, it is estimated that around 4300 HA and 900 HB patients are ...
